    Introduction About AMBISTRYN S 1GM INJECTION

    AMBISTRYN S 1GM INJECTION contains Streptomycin which belongs to the group of medicines called Aminoglycosides. AMBISTRYN S 1GM INJECTION is used to manage moderate to severe infections caused by susceptible strains of bacteria which includes tuberculosis infection (caused by Mycobacterium tuberculosis bacteria) and non-tuberculosis infections (caused by Pasteurella pestis/ tularensis, brucella, H. ducreyi, Klebsiella granulomatis, and/or H. influenzae bacterium) in affected individuals.

    AMBISTRYN S 1GM INJECTION should not be received by patients allergic to Streptomycin, and/or other aminoglycosides.

    Before receiving AMBISTRYN S 1GM INJECTION, inform your doctor if you have cranial (skull) nerve problems, vestibular nerve (responsible for motion and position) impairment, severe liver disease, and/or kidney problems (such as impaired kidney function, renal insufficiency, prerenal azotemia).

    AMBISTRYN S 1GM INJECTION is generally not recommended for use during pregnancy and in women planning for pregnancy since the medicine may cause ototoxicity (damage to the hearing/balance organs) in foetus. The medicine is advised to be received with caution by breast feeding women since the medicine may pass through breast milk.

    AMBISTRYN S 1GM INJECTION should be administered with caution in children and adolescents (aged below 18 years) and is advised to be received with caution by elderly patients (aged 59-years and above) after consulting the doctor.

    The most common side effects of receiving AMBISTRYN S 1GM INJECTION are vestibular ototoxicity (associated with symptoms such as vomiting, nausea, internal spinning sensation), paraesthesia (prickling/tingling sensation) of face, fever, rash, red itchy welts, and/or eosinophilia (associated with symptoms such as rash, itching, asthma). Consult your doctor if any of these symptoms worsens.

    How AMBISTRYN S 1GM INJECTION Works

    AMBISTRYN S 1GM INJECTION works by binding to 30S and 50S ribosomes (intracellular structures made of RNA and protein in bacterial cell) and inhibits protein synthesis in bacterial cell which leads to errors in the transcription of the genetic code resulting in lysis/destruction of bacterial cell wall thus managing and managing the spread of infection in affected individuals.

    How to use AMBISTRYN S 1GM INJECTION

    AMBISTRYN S 1GM INJECTION will be given to you only by a doctor or a nurse into a muscle as an intramuscular injection, or into a vein as an intravenous injection/infusion. Your doctor will decide the correct dose and duration for you depending upon your age, body weight and disease condition.

    Side Effects Of AMBISTRYN S 1GM INJECTION

    Common

    • vestibular ototoxicity (associated with symptoms such as vomiting, nausea, internal spinning sensation)
    • paraesthesia (prickling/tingling sensation) of face
    • fever
    • rash, red itchy welts
    • signs of eosinophilia (condition marked by increase in white blood cell) such as rash, itching, asthma

    Uncommon

    • deafness
    • exfoliative dermatitis (severe inflammation of entire skin surface)
    • signs of anaphylaxis (severe, life-threatening allergic reaction) such as skin rash, nausea, vomiting, difficulty breathing, and/or shock
    • haemolytic anaemia, leukopenia (low white blood cell count), thrombocytopenia (low platelet level), pancytopenia (decreased of all three blood cells),
    • muscular weakness
    • amblyopia (reduced vision in one eye)

    Rare

    Stop receiving AMBISTRYN S 1GM INJECTION and consult your doctor immediately if you experience any of the following side effects:

    • complete/partial deafness (loss of hearing)
    • signs of allergic reactions such as rash, hives, swelling of face, lips, tongue, throat, and/or difficulty breathing/swallowing
    • signs of kidney problems such as infrequent/painful urination, dark amber/red coloured urine, fatigue, muscle weakness, swelling, nausea, vomiting, and/or loss of appetite

    How To Manage Side Effects

    Nausea and vomiting:

    Receive AMBISTRYN S 1GM INJECTION with/just after a meal. Stick to simple meals. Avoid eating oil rich/spicy foods, sugary snacks and drinks. Avoid drinking too much liquid along with your meals, and drink slowly in between meals. Consult your doctor if the condition worsens.

    Itchy Skin:

    Try to pat or tap your skin instead of scratching it. Hold a damp towel to cool your skin. Wear loose clothing and avoid tight clothes, or clothes made from wool or synthetic fabrics. Avoid using perfumed soaps, deodorants or moisturizers and use a skin softener regularly.

    Warning & Precautions

    Pregnancy

    Do not drive or operate machines if you experience vestibular toxicity (associated with symptoms such as nausea, vomiting, internal spinning sensation) after receiving AMBISTRYN S 1GM INJECTION.

    Kidney

    AMBISTRYN S 1GM INJECTION should be used with caution in patients with kidney problems such as impaired kidney function, renal insufficiency, prerenal azotemia (type of kidney failure). Consult your doctor before receiving the medicine.

    Liver

    AMBISTRYN S 1GM INJECTION should be used with caution in patients with severe liver disease. Consult your doctor before receiving AMBISTRYN S 1GM INJECTION.

    Allergy

    Do not receive AMBISTRYN S 1GM INJECTION if you are allergic to Streptomycin, other aminoglycosides, and/or any other ingredients of this medicine.

    Others

    Before receiving AMBISTRYN S 1GM INJECTION inform your doctor if you:

    • have cranial (skull) nerve problems
    • have vestibular nerve (responsible for motion and position) impairment
    • recently undergone surgery/been under anaesthesia

    Use in Paediatrics:

    Interactions

    A. Drug-Drug interactions:

    Before receiving AMBISTRYN S 1GM INJECTION, inform your doctor if you are taking any of the following medicine:

    • antibiotics (used to manage/prevent bacterial infections) Ex. neomycin, kanamycin, gentamicin, cephaloridine, paromomycin, viomycin, polymyxin B, colistin, tobramycin
    • other antituberculosis medicines having ototoxic and nephrotoxic potential (Ex. capreomycin, viomycin, vancomycin
    • amphotericin B (a medicine used for management of fungal infections)
    • cephalosporin (used to manage wide range of bacterial infections) Ex. cefazolin, cefepime
    • cyclosporin (used to manage rejection of organ transplantation)
    • cisplatin (used to manage advanced cancer of bladder, ovaries, and/or testicles)
    • furosemide, etacrynic acid (medicines used to manage oedema and blood pressure)
    • neuromuscular blocking agents (used during surgery to manage muscle movement) Ex. rocuronium, vecuronium
    • zalcitabine (a medicine used for management in HIV patients)
    • mannitol (used to manage kidney failure, cerebral oedema)

    Overdosage:

    AMBISTRYN S 1GM INJECTION will be administered to you only by a doctor or a nurse in a hospital, and so it is unlikely to receive an overdose. However, consult your doctor or nurse if you experience any unusual symptoms such as nephrotoxicity (rapid deterioration of kidney function), ototoxicity (hearing/balance problems), neuromuscular blockage (blockade of skeletal muscles), and/or respiratory paralysis (complete/severe weakness of muscles of respiration).
