INTRODUCTION

MACIKAD 10MG contains Macitentan which belongs to the class of medicine called endothelin receptor antagonists. It is indicated for long-term treatment of pulmonary arterial hypertension (PAH) in adults. PAH is a type of high blood pressure that affects pulmonary arteries (blood vessels carrying blood from heart to lungs) which results in narrowing of the vessel walls, shortness of breath, dizziness and chest pain.

Macitentan widens your pulmonary arteries and makes it easier for the heart to pump blood through them. This in turn reduces blood pressure and relieves the symptoms of PAH. MACIKAD 10MG can be either used alone or in combination with other medicines to treat PAH. Your doctor will take several blood tests before and during the course of treatment to check whether you are anemic and to ensure whether your liver is functioning properly.

Take MACIKAD 10MG as directed by your physician. The recommended daily dosage is one tablet/day. It can be taken with or without food. The most common side effects of this medicine are headache, hypotension, anemia, liver dysfunction and respiratory disorders. Contact your doctor if any of the side effect bothers you. Before taking this medicine, inform your doctor if you have any kidney or liver disease. It should be used with caution in patients with pulmonary veno-occlusive disease, because the use of macitentan may lead to pulmonary edema. This medicine is generally not recommended for use in children (below 18 years) and in pregnant or breastfeeding women. Consult your doctor before taking this medicine.

HOW MACIKAD 10MG WORKS

Macitentan binds to and antagonizes the effects of endothelin receptors (ET-A and ET-B) present in the endothelial cells of blood vessels and smooth muscle. This prevents vasoconstriction of arteries, lowers blood pressure, relieves the symptoms of PAH and improves the course of the disease.

DIRECTIONS FOR USE

Take MACIKAD 10MG as directed by your physician. The recommended daily dosage is one tablet/day. It can be taken with or without food. Swallow the medicine as whole with a sip of water. Do not crush or chew the medicine. Your doctor will decide the correct dose and duration for you depending upon your condition. Continue to take this medicine, as long as your doctor tells you to do so, to obtain better results.

SIDE EFFECTS OF MACIKAD 10MG

COMMON

  • headache
  • low blood pressure
  • bronchitis (inflammation of airways)
  • nasopharyngitis (inflammation of throat and nasal passages)
  • pharyngitis (inflammation of throat)
  • nasal congestion
  • swelling of ankles and feet
  • influenza (flu)
  • urinary tract infection
  • leukopenia (low white blood cell counts)
  • thrombocytopenia (low blood platelet counts)
  • signs of anemia (such as dizziness, weakness, fast heart rate, pallor)
  • signs of liver disease (such as nausea, vomiting, stomach pain, jaundice, dark-colored urine, itching, lethargy, joint and muscle pain with fever)

HOW TO MANAGE SIDE EFFECTS

Nausea or vomiting:

Try taking this medicine with, or just after meals. Stick to simple meals. Do not eat rich or spicy food.

Stomach pain:

Try to rest and relax. You can eat and drink slowly or have smaller and frequent meals. Keeping a heat pad on your stomach may also help. If still pain persists, speak to your doctor.

Headache:  

Apply hot or cold-water bag on your head. Take rest in a quiet and dark room. Drink tea or coffee.

PREGNANCY

MACIKAD 10MG is not recommended for use in pregnant women unless necessary. Discuss the risks and benefits with your doctor.

BREASTFEEDING

MACIKAD 10MG is not recommended for use in breastfeeding women unless necessary. Discuss the risks and benefits with your doctor.

DRIVING AND USING MACHINES

Do not drive or operate any tools or machines, because this medicine may cause headaches and hypotension.

ALCOHOL

Avoid consumption of alcohol while taking this medicine.

KIDNEY

MACIKAD 10MG should be taken with caution in patients with severe kidney disease. Consult your doctor for advice.

LIVER

Do not take MACIKAD 10MG if you have severe liver disease. Consult your doctor for advice before taking.

ALLERGY

Do not take this medicine if you are allergic (hypersensitive) to macitentan.

LUNGS

MACIKAD 10MG should be taken with caution in patients with pulmonary veno-occlusive disease (obstruction of the lung veins). Consult your doctor for advice.

INTERACTIONS

Talk to your doctor, if you are taking,

  • medicines used to treat tuberculosis (e.g. rifampicin, clarithromycin, telithromycin)
  • medicines used to treat epilepsy (e.g. phenytoin or carbamazepine)
  • medicines used to treat depression (e.g. carbamazepine, nefazodone or St. John’s Wort)
  • ritonavir and saquinavir (used to treat HIV infections)
  • medicines used against fungal infections (ketoconazole (except shampoo), itraconazole or voriconazole)
